催乳素受體（PRLR）是單次跨膜受體，屬於Ⅰ型細胞激素受體超家族，含有兩個纖維連接蛋白Ⅲ型結構域。所有 1 類配體透過聚集機制活化各自的受體。配體結合導致跨膜PRLR二聚化，進而導致參與訊號路徑的分子（如Jak-STAT、Ras/Raf/MAPK）磷酸化和活化。 PRLR 不含有內在的酪胺酸激酶胞質結構域，但與胞質酪胺酸激酶 JAK2 有關。 PRLR 主要作為腦下垂體激素催乳素 (PRL) 的受體，PRL 是一種影響脊椎動物生殖和體內平衡的分泌激素。 PRLR 可以透過兩種不同機制的相互作用進行調節，即 PRL 或卵巢類固醇激素，可以獨立調節，也可以以組織特異性的方式組合調節。荷爾蒙催乳素 (PRL) 在乳癌發病機制中的作用是由其同源受體 (PRLR) 介導的。負向調節 PRL 訊號傳導的 PRLR 的泛素依賴性降解是由 PRL 介導的 PRLR 在 Ser349 上的磷酸化引發的，隨後募集含有 β-轉導蛋白重複序列的蛋白質 (β-TrCP) 泛素-蛋白質異肽連接酶。 PRLR穩定性的改變可能直接影響乳癌的發病機轉。
