程序性死亡-1（PD-1）先從小鼠T細胞雜交瘤2B4.11克隆出來，是CD28/CTLA-4超家族成員之一。 PD-1屬於I型跨膜蛋白，是重要的免疫抑制分子。 PD-1 的胞質尾含有兩個結構基序，一個基於免疫受體酪胺酸的抑制基序 (ITIM) 和一個由兩個酪胺酸殘基形成的基於免疫受體酪胺酸的轉換基序 (ITSM)，這兩個酪胺酸殘基決定了 PD-1 訊號介導的差異。小鼠 PD-1 在胸腺中表達，與人類 PD-1 有約 69% 的 aa 序列同一性。最近發現，程序性死亡-1 (PD-1) 及其配體，程序性死亡配體 B7H1 (PD-L1) 和 B7DC (PD-L2)，在與配體結合後可調節 T 細胞活化和耐受性，以抗原特異性的方式抑制 T 細胞效應功能。 PD-1基因敲除小鼠會誘發一些自體免疫疾病，提示PD-1作為共抑制分子，積極參與維持週邊耐受性。因此，PD-1可能成為癌症、感染、自體免疫疾病以及器官移植的免疫療法的有用標靶。
